There was a time when to amplify DNA,
You had to grow tons and tons of tiny cells.
Then along came a guy named Dr. Kary Mullis*,
Said you can amplify in vitro just as well.
Just mix your template with a buffer and some primers,
Nucleotides and polymerases, too.
Denaturing, annealing, and extending.
Well it's amazing what heating and cooling and heating will do.
PCR, when you need to detect mutations.
PCR, when you need to recombine.
PCR, when you need to find out who the daddy is.
PCR, when you need to solve a crime.
*Kary Mullis，1993 年因发明了PCR技术获得诺贝尔化学奖。
要翻译过来，大概就是这么说的：
以前如果要扩增DNA，就得培养很多很多的那小小的细胞（细胞带有目的DNA，属于体内扩增)。
后来有个叫Kary Mullis 的家伙跑出来说，其实你可以在体外很简单的达到目的。
只要把你的模板DNA 跟缓冲液混匀了，当然还要加一些引物，核苷和聚合酶。
然后，变性（94 度，让DNA 双链变性成单链）、退火（55 度左右，让引物可以跟匹配的DNA 序列结合）、延伸（72 度，聚合酶的工作温度，目的DNA 片断被复制），你会发现加加热降降温也有奇妙的效果。
当你想检测突变时，来做PCR 吧；
当你想重组基因时，来做PCR 吧；
当你要亲子鉴定时，来做PCR 吧；
当你要解决案件时，来做PCR 吧。
